Role of dendirtic cells in crescentic glomerulonephritis

Kidney dendritic cells (DCs) and macrophages (MF) are central regulators in crescentic glomerulonephritis (cGN) and in other types of kidney disease. Their functionality is affected by numerous factors, such as cytokines, chemokines and complement components. We previously showed that the chemokine receptor CX3CR1 specifically recruits DCs into the renal cortex, where they drove progression of cGN. In the last funding period, we discovered that a) the only CX3CR1 ligand, CX3CL1, is ectopically upregulated in the medulla and cortex of the inflamed kidney, b) that loss of CX3CL1 attenuates cGN, c) generated mice lacking complement receptors (CR) 3 and 4 and studied the effects of complement on hypertensive kidney disease, and d) established techniques for single cell transcriptome analysis of renal DC/MF. In the next funding period, we would like to address the following topics: 1) Role of ectopic CX3CL1 expression in repositioning of renal DC/MF within the inflamed kidney in cGN, 2) CX3CL1 expression as a diagnostic parameter in biopsies of kidney disease patients. 3) Role of CR3/4 in DC activation in cGN. 4) Transcriptomic profiling of intrarenal single DCs/MFs during cGN and alignment with data from patients with cGN. Our long-term aim is to obtain a better understanding of renal DC/MF in cGN and identify targets for therapeutic strategies.
